课题名称 新疆蜱传原虫病分子免疫流行病学研究 Immunologic and Molecular epidemic study on tick- borne Protozoan disease in Yaks in Xinjiang province 执行时间 2010年 1 月 -2012年 12 月 课题编号 30960282 项目来源 国家自然科学基金委 资助经费 23 万 主 持 人 巴音查汗 结 题 否 是 成 员 岳成、郭庆勇、米克热木、赵红琼、张晓红、王振宝、王常汉、杨帆 摘 要 新疆具有养殖牦牛的广阔高山草原,牦牛又是我区特色畜种,由于新疆境内分布的蜱种类约占全国的2/3,而其引起的蜱传原虫病长期制约牦牛业的发展,阻碍高原牧区人们脱贫致富之路。目前,对蜱传原虫病的诊断仍用镜检法,由于循环血染虫率低或相似虫体鉴别难等原因,易造成漏诊和误诊,而血清学诊断法存在着抗原敏感、特异性差等问题。本项目通过运用现代分子免疫学检测法来弥补常规检查法,以《巴州牦牛疫病防治研究》和日本《高级原虫病研究》为前期工作基础,调查研究媒介蜱及蜱传原虫病。对牦牛泰勒虫、巴贝斯虫病进行DNA和基因型分析,建立地方虫株体外培养技术。优化血清学诊断方法,引用PCR和实时荧光定量PCR技术,从而提高检出率。鉴定媒介蜱,全面了解蜱传原虫病的感染率、感染强度、流行特点等,以此流行病学参数正确估计危害,阐明防治重点,拟定综合防治规划。为建立分子免疫诊断体系和综合防治牦牛疫病提供科学依据和技术支撑。 Abstract Xinjiang has a wide alpine steppe to raise poephogus grunniens which are the livestock with special featurein in the local region.As the category of the tick distributed over xinjiang Province account for about two thirds of the natioi-wide one.However, the protozoiasis transmissed by tick the people in the plateau farming area get out of poverty.Now, the diagnosis for the protozoiasis transmissed by tick still use microscopic examination method because of the several reasons as follows:the low rate of circulating blood-staining worm or the difficulty in distinguishing the similar polypide may lead to polypideand missed diagnosis and misdiagnosis,and there exist some problems such as antigen sensitiveness and bad specificity in the serological diagnosis method.This project investigate and discuss intermediary tick and the protozoiasis transmissed by tick by the contemporary molecular immunology detection method to make up routine examination on the basis of 《the prevention and cure and research for Bazhou poephogus grunniens plague》and《the senior protozoiasis research in Japen》.We analyze genotype of the Theiloriosis and Babesiosis , establishing culture in vitro technology for the local worm strain. We optimize serological diagnosis, by means of real-time PCR and PCR technology thus improving the detection rate..We identify intermediary tick to have a overall knowledge of the epidemiology parameters for protozoiasis transmissed by tick such as infection rate and infection magnitude and the ;epidemic characteristic by which estimate harm exactly, illuminatingthe point of prevention and cure, laying integrated control program ,which provide scientific theory and technology support to establish the system of molecule immunodiagnosis and integrate control for poephogus grunniens plague.
